   This research focuses on engineered extracellular vesicles (EVs) for concurrent immunotherapy and chemotherapy targeting cancer. The study developed EVs that overexpress high-affinity variant PD-1 protein (havPD-1) while simultaneously knocking out PD-L1 and beta-2 microglobulin in donor cells. These engineered havPD-1 EVs demonstrated multiple therapeutic effects: they reduced PD-L1+ cancer cell proliferation, induced cellular apoptosis, efficiently blocked PD-L1 mediated T cell suppression, and showed robust anti-tumor activity in xenograft models. Furthermore, the researchers enhanced therapeutic efficacy by loading the EVs with PARP inhibitors, creating a dual-action delivery system that combines immune checkpoint inhibition with targeted chemotherapy.
